Since Kishimoto has nelected to pull a Trekkie on us and explain what's behind the Curse Seals, I have some theories to share.
The exact cause of the Curse Seals's profound effects is a unique enzyme that Juugo's body naturally produces in large amounts. Orochimaru must have extracted this enzyme (obviously) to make the curse seals.
Juugo
Clearly since Juugo is the source material, he would have a 100% concentrated, pure enzyme flowing freely in his body (which would of course be why Juugo is so very powerful with it; The only reason Sasuke is more powerful is because he is heavily aided and abetted by Orochimaru's power and his own bloodline).
My theory is that perhaps Orochimaru was unable to extract the enzyme in its pure essence, meaning that Juugo's DNA was still lingering within the enzymes (which would explain why the recipients' bodies rejected the enzyme so badly, because it was foreign DNA.
The Enzyme Itself
I believe that the Seal may actually act as a surrogate 'Gland' from which the enzyme is produced, which over time absorbs small amounts of chakra from the host to greatly accelerate the chemical reactions within the body, i.e. chakra production, physical strength, etc, the chakra being the primary catalyst for the transformations. Granted, because the enzyme is combined with Orochimaru's chakra (which allows for him to control it more easily) which provides the 'dark' element of the chakra.
The first four curse seals (Soul, Mind, Body, and Jirobo's Seal) I would image would be of relatively low concentrations (Jirobo's being of the lowest concentration at perhaps around 50%, Soul being the highest at around perhaps 70%), while the Earth Seal might be at 80-85%, being much more powerful than the others.
The Heaven Seal would be even more powerful still, around maybe 95% (hence Sasuke's immense power with it and the most extreme transformation).
Conclusion?
Theoretically, might it be possible to forcibly separate the enzyme from Juugo's DNA (which would both purify the enzyme and eliminate the biological rejection), and then using purified chakra supersaturate it so that it could acheive an even higher concentration, thus producing a more potent transformation?
From there, the concentrated enzyme (anywhere from 110% to 150% concentration) could potentially be saturated with more chakra from an external source for control, or introduced 'straight-up'.
Good theory or no?

Makes sense to me and proves what I had already thought, in regards to both Juugo and the combination of that perverted annoyance's chakra. The resulting hypothesis also makes sense to me.

Theoretacally solid. Yet until Kishimoto says otherwise your theory has the best possibilities for and advance(if not extention) the naruto story plot even more.(IE Kabuto with the most powerful curse seal now thats a twist)

And bluen, your analysis intrigues me as well...
This interesting little twist would prove VERY interesting if I were to incorporate it into my little disease that I shall soon make public...
Perhaps I shall even ask Orochimaru himself (he's back, right?) if he would like to instigate it... perhaps even create it...

Your theory seems solid, and yet, it is just that, theory.
I believe your conclusion, Loxagn, conflicts with your earlier statement about Juugo's DNA conflicting with the DNA of another, even if you tried a supersaturate of chakra. I believe if anything, Orochimaru tried it already. I mean, he is after all, one of the best kinjutsu users seen in the series. If he has failed, then I believe anyone else would fail.
And also, Orochimaru has extracted the enzyme from Juugo which gives us the present-day curse seal, so forcibly extracting the enzyme would do no more than what Orochimaru has done.
And let us not forget, that it is within Juugo alone who has the power to utilize the power of the seal without an actual seal. Set forth by the data books, the actual seal is a Ninjutsu/Fuuinjutsu, only known by Orochimaru. This is backed up by the fact that only he is able to place the seal on others, because he is the only one that knows the jutsu. So even if you were able to isolate the enzyme yourself, you'd have no way to draw the power out of it unless through some kind of seal, and only Orochimaru would know how.
I do not see a way to avoid the negative effects of the curse seal, no matter how you try to mess with DNA simply because you can not combine non-compatible DNA. However, there might be a way to increase the power of the seal through something Loxagn was talking about, but in doing so I believe that it would also increase the rate of the negative effects as well.
The only way I see in naturally and potentially lessening the dangers of the seal while possibly increasing the power, is to either find some one with the same traits as Juugo and make them mate, or engineer test-tube children with Juugo's genes and other kekke genkai until you get one that works.
Anyway, my two-cents on it.

as captain jack would say "how about i add a machete to your intellectual thicket?"
i agree with the theory. but let me add a new angle to this, Juugo, uses the enzyme in his cells to convert molecules into a ... well ATP on steroids. ((that is what i gather from your theory and what i know about enzymes)) but then there is somthing that juugo's body does that prevents him from .... deteriorating like the rest of the people. true it may be because the DNA is uncompatable ... or it is there is not one but two sets of enzymes in his body one to convert the molecules into the powerful energy, and one to convert that acts as a filter one that doesnt harm him ((btw i am only assuming that he doesnt get hurt.))
now for those of you who are car savvy you can think of this like a catalitic converter in a car, it will limit down the power so you dont blow up the engine.
now, i would say that the heaven seal is more uf an unrefined way, almost as if the second enzyme has been removed, thus hurting the user, but giving him more power as a result. ((this would explain why the heaven seal has such power.)) so in theory there would be a way to restrict the effects of the curse seal by studying the second enzyme. ((but that is if we take the enzyme approach.))
another way to think of the curse seal as a virus that has been mutated by something specific in Juugo's body. Virus would explain how it can be passed to anyone, and why it takes time to spread about the body. so, in theory there should be a way to synthesize an antibody to ... well cure the curse seal, or at least nullify the affects.
hmmm? thoughts?

I agree with Haku on this one, BUT I do have one thing to add. Blood transfusions and organ donations are based on similar things. In order for the body to accept a donated organ would be for it to match your body, but since I don't know the entire science behind organ donations, I'll stick with blood because I can explain what is in my mind that way. In order for a blood transfusion to be successful, the blood must match the bodies type(A B AB or "O") respectively. I think the same could be said about the curse seals.
