When I'm hiring (wink-wink at Ritz xD) I usually look for whoever sticks around in the shop AFTER posting an application. Once, I had a shop called Enchanted Glade. I hired a few people I thought were best for the job--until the shop opened, and they were nowhere to be found. No PM's were answered, etc. I also look for literacy, because regulars have to be able to understand them. xD
When you look at some of these interest threads, hiring tons of people, I laugh (like a PSYCHO-MANIAC! >D ) because, seriously, do you need 8 colorists? Is it really essential to have 25472385748759423 managers? No! ONE person can handle guild stuff. ONE person can take care of moderating the thread, as well as updating it. NOT twenty. These things usually only take twenty minutes at most--not twenty people. And if colorists can't be busy with work, then why bother applying? They know that that's what they're in for, so you don't need to hire forty staff members.
Ranting aside, Ritz, the most a shop should have is 5 staff members. 8-9 if the shop gets really popular.

That's going to depend a lot upon what type of shop you have and the workload of each individual.
There is no hard and fast number of staff that is optimal for every shop, though when first starting out, a core team of 1-3 is good. The more part-time and guest coloring staff you have, the more total number of staff - if you have mostly F/T staff putting out many pets each, then you will have a smaller number.
The complexity of the pets themselves also determines your staffing needs - the more involved the base art, the more edits and details, and the more stages, the longer each pet will take to do.
